چکیده
Pro-inflammatory Interleukin-1 receptor antagonist (IL-1RN) gene polymorphism and high risk Helicobacter pylori virulence factors in combination plays an important role in the outcome of H. pylori associated diseases, which can even lead to gastric cancer. The purpose of this study was to analyze the prevalence of high risk host and bacterial factors such as IL-1RN gene variable number tandem repeat (VNTR) polymorphism and cagA, vacA and iceA genotypes among a group of H. pylori infected dyspepsia patients from Kozhikode area of Kerala state, India. Mucosal biopsies from fourty two randomly selected dyspepsia patients infected with H. pylori, as determined by rapid urease test (RUT) and glmM gene specific PCR, were used in this study. The high risk bacterial virulence factors such as vacA,cagA,iceA1 and iceA2 genotype status of colonized H. pylori and the high risk host proinflammatory IL-1RN*2/2* allele status were examined by PCR methods. The high risk vacAs1/cagA/IL1RN*2/*2 genotype combination was found in six patients (14%), vacA s1/cagA/IL-1RN*1/*1 in three patients (7%) and vacAs1/cagA/IL-1RN*1/*2 in seven patients (16%). In addition, analysis of allelic variation of iceA gene of H. pylori, such as iceA1 and iceA2 genotype, in colonized patients has revealed the co-existence of both iceA1 and iceA2 strains among 31% of patients included in this study.

Helicobacter pylori and interleukin 1 genotyping: an opportunity to identify high-risk individuals for gastric carcinoma.
BACKGROUND Both Helicobacter pylori genotype and host genetic polymorphisms play a role in determining the clinical consequences of H. pylori infection. We investigated whether there are any combinations of bacterial and host genotypes that are particularly associated with the occurrence of gastric carcinoma. METHODS Genotypic variations in virulence-associated genes of H. pylori vacA (s and ...

BACKGROUND AND OBJECTIVES Helicobacter pylori has been strongly associated with peptic ulcer diseases, chronic gastritis, ulcers, and reported as a risk factor for gastric cancer, too. The vaculating cytotoxin (vacA), the cytotoxin associated genes (cagA), the induced by contact with epithelium factor antigen (iceA gene), blood adhesion binding antigen (babA2), and outer membrane protein oipA h...

The clinical outcome of Helicobacter pylori infection has been particularly associated with virulence genotypes. These genotypes are useful as molecular markers in the identification of patients that are infected and at high risk for developing more severe gastric pathologies. Our main objective was to determine the prevalence of virulence genotypes cagA, vacA, iceA and babA2 of H. pylori, in p...
متن کاملPrevalence of Helicobacter pylori vacA, cagA and iceA genotypes in Nigerian patients with duodenal ulcer disease.
Distinct virulence factors of Helicobacter pylori have been associated with clinical outcome of the infection; however, considerable variations have been reported from different geographic regions. Data on genotypes of African H. pylori isolates are sparse. The aim of this study was to determine the prevalence of specific genotypes of H. pylori in Nigerian patients with duodenal ulcer and non-u...

Infection with Helicobacter pylori strains harboring determinants of pathogenicity may lead to a strong inflammatory response in gastric mucosa. In this work, we examined the frequency of the cagA, vacA and iceA genotypes in H. pylori strains isolated from Brazilian patients and correlated these with the clinical manifestations. H. pylori was isolated from 165 patients [30 with non-ulcer dyspep...
